Flow Cytometry Protocols

Krishan Stain for DNA Cell Cycle Analysis

  1. Prepare a single cell suspension with 0.1 to 1 x 10-6 cells per tube
  2. Wash cells with PBS, then centrifuge to pellet them
  3. Remove the supernatant PBS
  4. Vortex to dislodge the pellet
  5. Add 1 ml of Krishan’s stain
  6. Vortex again
  7. Refrigerate and let stand at least overnight before analysis

Saponin/PI Stain for Apoptosis & DNA Cell Cycle Analysis

  1. Prepare a single cell suspension with 0.1 to 1.0 x 10-6 cells per tube
  2. Wash cells with PBS, then centrifuge to pellet them
  3. Remove the supernatant PBS
  4. Vortex to dislodge the pellet
  5. Add 1 ml of saponin/PI stain Saponin PI stain
  6. Vortex again
  7. Refrigerate and let stand for 6 to 24 hours before analysis, but no longer than 24 hours
